Spence scored on a 22-yard run at 6:37 in the fourth quarter to set the Pioneers (9-2, 3-2 Northeast Conference) up for a showdown next week against conference leader Robert Morris.
Spence carried the ball 35 times and earned the lion's share of Sacred Hearts 299 total rushing yards, compared with 113 for Duquesne (5-4, 3-2 Northeast Conference).
Sacred Heart's Troy Moore sealed the game with an interception late in the fourth quarter off a Dillon Buechel pass, bringing to 20 the number of pick-offs by Pioneers defenders this season.
